We usually take measurements using pins and then back work the results into software to obtain the circular tooth and space widths.
We also use measurements obtained by the Base Tangent Length or Span technique although this is not as accurate as using pins.
Measurements taken directly at the pitch line, on external splines only, are usually taken with gear tooth verniers resulting in chordal dimensions which have to be converted back to circular dimensions.
Hexagon WN8 should be able to do all the math for you.
